Abstract

Early detection of cancer is vital for improving the survival rates, but due to limitations including cost and invasiveness, frequent screening for cancer is not viable. On the other hand, biosensors can be offered as an alternative solution. Liquid biopsy is a less invasive method for obtaining biological fluids as biosensor samples, while biosensors can detect specific biomarkers in these samples. EVs can serve as potential biomarkers for cancer detection due to their roles in cancer development. In this project, the EVs were isolated from A549 cell lines via sucrose cushion ultracentrifugation, and later characterized and quantified by TEM, dot blot, NTA, and BCA assay, which fulfilled the MISEV 2018 guidelines. The developed biosensor in this project consisted of the ternary composite AuNP/MOF/PEDOT:PSS as the sensing material, and CD63 aptamer as the bioreceptor. The ternary composite AuNP/MOF/PEDOT:PSS was also characterized by FESEM, EDX, FT-IR, XRD, XPS, CV, and EIS. Lastly, the performance of the developed biosensor under optimized conditions was evaluated across several parameters, including linear range, LOD, reproducibility, stability, selectivity, and clinical applicability.
